Relation Between Culture Medium, Hormones Combination and Shoots Proliferation of Plum Cv. “Santa Rosa”

Abstract

Background and Objective: Plum produced all over the world. Plum fruit has high nutrient value (these fruits are rich in vitamins, minerals and proteins). In vitro shoot tips of plum (Prunus Salicina) cv."Santa Rosa" was used to investigate the effect of some growth regulators on multiplication and rooting. Materials and Methods: Benzylaminopurine (BAP) at different concentrations (0.00, 0.25, 0.50 and 1.00 mg L1) was studied for their ability to induce plum shoot multiplication. In addition, one and half, full and double strength of MS medium were investigated. Moreover, different types of auxin (NAA, IBA and IAA) with two concentrations (0.25 and 0.50 mg L1) alone or in combination with 1.0 mg L1 Activated Charcoal (AC) were tested for rooting process. Results: The obtained results revealed that BAP affected shoot multiplication positively as (0.5 mg L1) increased shoot number followed by 1.0 mg L1 compared with the control. However, the higher concentration used, the more vitrification achieved. Double and one and half strengths of MS medium obviously enhanced shoot number, shoot length, leaf number and chlorophyll. The highest root percentage was observed with full strength MS supplemented with 0.25 mg L1 IBA. Conclusion: Using BA at low concentration was recommended to improve shoot multiplication rate meanwhile high levels of BA resulted in vitrification phenomena. Although, in some cases, raising culture medium strength is required to face increasing nutrients needs for high shoot multiplication rate for several plants, however in plum cv (Santa Rosa) full strength of MS was recommended for high multiplication rate of both of shoots and roots.
